A. E. Carlsson Department of Physics, Washington University, St. Louis, Mo. 63130 (Dated: February 2, 2008) Abstract Theory and experiment have established that F-actin filaments are strongly attached to the intracellular parasites (such as Listeria) they propel with “comet tails”. We consider the implications of these observations for propulsion. By calculating the motion produced in various models of attachment and comparing to experiment we demonstrate that the attachment must be sliding rather than hinged. By modeling experiments on ActA-coated spheres we draw conclusions regarding the interaction between F-actin and their surfaces that may also be applicable to living systems.

How VASP enhances actin-based motility
The function of vasodilator-stimulated phosphoprotein (VASP) in motility is analyzed using a biomimetic motility assay in which ActA-coated microspheres propel themselves in a medium containing actin, the Arp2/3 complex, and three regulatory proteins in the absence or presence of VASP. Propulsion is linked to cycles of filament barbed end attachment-branching-detachment-growth in which the ActA...
متن کاملA biomimetic motility assay provides insight into the mechanism of actin-based motility
Abiomimetic motility assay is used to analyze the mechanism of force production by site-directed polymerization of actin. Polystyrene microspheres, functionalized in a controlled fashion by the N-WASP protein, the ubiquitous activator of Arp2/3 complex, undergo actin-based propulsion in a medium that consists of five pure proteins. We have analyzed the dependence of velocity on N-WASP surface d...
متن کاملActin Depolymerizing Factor (ADF/Cofilin) Enhances the Rate of Filament Turnover: Implication in Actin-based Motility
Actin-binding proteins of the actin depolymerizing factor (ADF)/cofilin family are thought to control actin-based motile processes. ADF1 from Arabidopsis thaliana appears to be a good model that is functionally similar to other members of the family. The function of ADF in actin dynamics has been examined using a combination of physical-chemical methods and actin-based motility assays, under ph...
متن کاملModeling the evolution of cells outgrowth due to the force exerted by actins
Motility and membrane deformation are crucial to motile cells. Therefore formation of protrusion in the membrane has been the subject of various studies. The stable shape of the membrane and also its movements are controlled by the forces exerted by actin filaments. In order to study the protrusion behavior, we represented a toy model based on actin filaments polar characteristic and elastic ch...
متن کاملVelocity oscillations in actin-based motility
We present a simple and generic theoretical description of actinbased motility, where polymerization of filaments maintains propulsion. The dynamics is driven by polymerization kinetics at the filaments’ free ends, crosslinking of the actin network, attachment and detachment of filaments to the obstacle interface and entropic forces. We show that spontaneous oscillations in the velocity emerge ...
